Biotechnological methods in the treatment of degenerative damaged cartilage of intervertebral disc

In this work the new model of degenerative damage of the intervertebral disc (IVD) in rats, that corresponds to the nature and mechanism of development of degenerative changes occurring in the human IVD compared to the traumatic way. The novelty of the model is confirmed by the issuance of the patent of Ukraine. Based on the analysis of the results of modeling degenerative damage of the IVD in rats it was found that the fixation of the caudal spine in a flexed position during 60 days leads to severe degenerative changes in the IVD CcVI-VII, the most pronounced component of which is the central focal necrosis of fibrochondrocytes with form of cracks in fibrous ring (FR).

In this series of works the two routes of administration of the cell suspension of cryopreserved multipotent mesenchymal stromal cells (cMMSCs) into IVD CcVI-VII were also compared. It was a widespread injection and designed by the author and patented method on a gelatin sponge (patent of Ukraine for useful model №89953). The method of administration of cMMSCs on a gelatin sponge provides significantly better results compared to injecting one.

In further studies, it was found that the efficiency of bone marrow cMMSCs is directly dependent on the dose of cells. In the line of processed doses the optimal result was observed in the case of using cMMSCs in the amount of 0.25×106 cells.

In the study of the dynamics of the recovery processes, it was found that cMMSCs, local administrated in the area of degenerative changes of the IVD in experimental compression moel, have a regenerative effect on damaged cartilage of FR with the restoration of the integrity of the collagen fibers, the height of FR, the numerical density of fibrochondrocytes and radiographic density of FR on the 90th day after therapy.

The results of the research are presented in 21publications, incl. 1chapter of the monograph and8 articles in refereed journals. The H-index is 4, from 2013 - 3 (according to the Google Academy data). The presented works are quoted in more than 17 scientific and applied scientific journals. Innovation and competitiveness of technical solutions are protected by the 3patents.
